What is pGlo?

What is the point of the heat shock and transformation solution?

• To kill bacteria we don't want
• To make the bacteria's membrane open so the pGlo can go in
• To heat the plasmid up so ligase will work
• To make the pGlo plasmid

How often do you need a new loop?

• You'll use the same one the entire lab
• You should switch every 5 minutes
• Use a new loop every time you touch new/different bacteria/plasmid
• It doesn't matter

How many times do you put the tubes on ice, and for how long?

• Once for 10 min
• Twice for 5 min each
• Once for 10 min, a second time for 2 min
• 5 min, then 2 min, then 5 min again

Where should you label your petri dishes?

• on the lid plate
• on the bottom plate 

How long should the tubes be in the 42^oC water bath?

• 30 seconds
• 50 seconds
• 60 seconds
• 120 seconds

When moving the tubes from water bath back to ice, you should

• take your time
• do this quickly
