• เป็นการเพาะเชื้อที่ให้ผลบวกจากขั้น confirmed test ลงในอาหารจ าเพาะเช่น Eosin

methylene blue (EMB) agar หรือ Endo agar แบคทีเรียที่สามารถหมักน ้าตาลแลคโตส
ได้จะเกิดโคโลนีที่มีสีเขียวมันวาวมีสีด าตรงกลาง

• น าโคโลนีดังกล่าวกับไปเลี้ยงในอาหาร Lauryl sulfate-tryptose-lactose บ่มที่อุณหภูมิ

35 oC เวลา 24-48 h และสังเกตการสร้างกรดและแก๊ส
